Background and aim : molecular makers are useful approach for recognition of mutation in p. vivax.The aim of this study was genetic analysis of the dihydrofolate reductase -thymidylate synthase gene in p.vivax in hormozgan province for mutation in five codons realated with antifolate drug resistance.Material & methods: from april 2007 through January 2008 , a total of 118 blood samples were collected From p. vivax materil patient in tow endemic areas of Hormozgan province south of iran PCR semi -nested -pcr , relp and sequencing methods were used. Dhfr gene of p. falciparum from 16 isolate of jask area were also analyzed by sequencing .results : out of seventy two p. vivax isolates from jask and 46 isolates from minab the pvdhfr allers were seen in 32 samples . Mutatin was found in 28 (23.72) and 8 (608 ) isolate in 58 117 and 57 codons respectivity . amoung 32 mutant isolate double mutation in the from of s58R-S117n and f 57l-triple mutations in codons 57 , 58 and 117 were observed in 3 (9.4 ) of the isolates the 59 and 108 codone were mutant in all of the isolates of dfr gene of plasmodium falaciparum.Conclusion : In spite of this fact that antifolate drugs are not prescribed for p . vivax malaria, observedMutant alleles in pvdhfr gene of p.vivax pyrimethamine drug . Since fansider is using as the first -line of Anti- falciparum malaria , the obtained mutant alleles in pfdhfr gene is expectable.
